Staining: Professionally stained with Hematoxylin and Eosin (H&E). The high density of developing T-cells (thymocytes) in the cortex results in a deep purple stain, while the medulla appears lighter.

Section Type: Thin section (5–7 microns) revealing the organ’s internal partitions and cellular organization.
Key Histological Features:

Capsule & Septa (Trabeculae): An outer connective tissue capsule that extends inward as septa, dividing the parenchyma into many incomplete lobules.

Cortex (Outer Darker Zone): Densely packed with immature T-cells (thymocytes). This is the site of rapid cell proliferation and the initial stages of T-cell maturation.

Medulla (Inner Lighter Zone): Contains fewer, more mature T-cells and specialized epithelial cells.

Hassall’s Corpuscles (Thymic Corpuscles): A diagnostic feature found only in the thymic medulla. These are concentric layers of flattened epithelial cells that appear as pink, “swirled” structures. * Blood-Thymus Barrier: While not visible as a single structure, the cortical capillaries are highly specialized to prevent circulating antigens from reaching developing T-cells.
