Staining: Professionally stained with Hematoxylin and Eosin (H&E). The dense clusters of lymphocytes (White Pulp) appear deep purple, while the blood-rich areas (Red Pulp) appear bright pink to red.

Quality Control: Selected for clear visualization of the splenic nodules and the surrounding venous sinuses.
Key Histological Features:

Capsule & Trabeculae: A thick outer layer of connective tissue with inward-extending “beams” (trabeculae) that provide structural support and carry blood vessels into the organ.

White Pulp: These appear as circular or elongated purple clusters. They consist of Periarteriolar Lymphoid Sheaths (PALS) and Splenic Nodules (follicles) rich in B-cells.

Central Arteriole: A characteristic feature of the White Pulp; a small artery often located eccentrically within the lymphoid tissue.

Red Pulp: The more abundant, pink-staining tissue. It consists of Splenic Cords (Cords of Billroth) and Splenic Sinusoids where blood is filtered.

Marginal Zone: The boundary between the red and white pulp, where many macrophages and dendritic cells are located to trap antigens from the blood.

Hemofiltration: The Red Pulp acts as a “quality control” center, identifying and removing old, damaged, or abnormal erythrocytes (red blood cells).
